Highly methylated DNA can be enriched greater than 40-fold. 2.5 μg of MCF-7 DNA was enriched and eluted in triplicate with the MethylMiner system. qPCR was used to assess the enrichment of the two PLAU amplicons shown in figure 2. Mass of eluted DNA was measured for each fraction with a microplate-based PicoGreen assay. Absolute fold enrichment was determined as described in the text. Amplicon 1 (blue bars) was enriched in several fractions, possibly due to variable methylation in this locus or neighboring sequences. In contrast, amplicon 2 (orange bars) was almost exclusively enriched, to greater than 40-fold, in the highest salt fraction.
