Chapter 9. Figure 17. Illustration of Stem-loop Rt-qPCR method. TaqMan-based real-time quantification of miRNAs involves two steps, stem-loop RT and real-time PCR. Stem-loop RT primers bind to the 3' end of the targeted miRNA through annealing and are reverse transcribed to make first-strand cDNA which includes the miRNA and reverse primer binding site. The purpose of the tailed forward primer is to increase the melting temperature if needed, depending on the sequence composition of the miRNA.
