Chapter 6. Figure 2. Emission spectra, and spectral overlap, of four commonly used fluorescent dyes (FAM, HEX, ROX, and Cy5). To deconvolute the signal of multiple dyes within a single channel, dye calibration information is used to remove all fluorescent-signal present in a given channel that does not belong to the dye of interest. In short, dye calibration information is used to subtract, using simultaneuous equations, any dye bleed-through emissions (denoted by an asterisk above) from the emission signal generated by the dye of interest.
