Chapter 1. Figure 3. Equilibrium and kinetic paradigms of PCR. PCR can be described by equilibrium (left) or kinetic (right) models. Both paradigms highlight three events which occur during the PCR cycle (denaturation, annealing and extension). The more conventional equilibrium paradigm focuses on a specific temperature at which a reaction occurs and its time period (e.g. extension at 72 C for 15 s). The transitions between specific temperatures are generally ignored. The kinetic paradigm, however, differs in that the temperature of the reaction is never static. Denaturation, annealing and extension occur over a range of temperatures and can occur simultaneously. Reaction rates depend on temperature but are not limited to a single temperature.
