Chapter 1. Figure 1. The PCR cycle. Initial template DNA is denatured by heat. The reaction is then cooled to anneal two oligonucleotide primers, each annealing to opposite strands with their 3'-ends pointed inward toward each other. A polymerase then extends each primed template to double the amount of targeted DNA. The cycle is repeated 20-40 times through successive steps of denaturation, annealing and extension. Eventually if reagents are not limiting, the concentration of accumulating products becomes high enough that double strand product formation is favored over primer annealing, and the reaction plateaus.
