Abstract

HIV vaccine developers increasingly believe that the best protection against HIV/AIDS will probably require both cellular immunity and antibodies that neutralize a broad range of viral strains. However, designing immunogens that elicit such antibodies has emerged as one of the most challenging problems facing the field. This article reviews these challenges, most of which pertain to the structural features of the HIV envelope (gp120 and gp41). It also describes several strategies being pursued to overcome them by favorably presenting conserved epitopes that have been identified as targets for broadly neutralizing antibodies.
