Chapter 9. Figure 11. 3 million Human 293Gt cells were fixed with 1% formaldehyde for 10 minutes at room temperature. Subsequently, cells were lysed in 150 microliters MAGnify Lysis Buffer (10 minutes on ice) prior to chromatin shearing. Samples were either unsonicated (lanes 2, 3) or sonicated (lanes 3, 4) for 12 cycles using Bioruptor from Diagenode. 10μl of DNA was analyzed on a 2% E-gel. Lane 1: 100bp Ladder. Lane 2: Unsonicated DNA. Lane 3: Unsonicated DNA. Lane 4: Sonicated DNA from 1million 293Gt Cells. Lane 5: Sonicated DNA from 1million 293Gt Cells.
